ONLINE COVER iCARs on Target. T cells with PD-1 and CTLA-4 based inhibitory chimeric antigen receptors (iCARs) preferentially eliminate the target cells (CD19+GFP+, green) while sparing the off-target cells (CD19+PSMA+mCherry+, red).This is a representative image from the 3 hour time point of 38 hours of time-lapse microscopy. In this week’s issue of Science Translational Medicine Fedorov et al. show that CAR T cells without the inhibitory receptor kill both populations, whereas the iCAR T cells only kill the target cells.
