Human disease is a complex network of interacting pathways. As such, it may be necessary to treat the system with a combination of drugs to boost efficacy and reduce side effects. In this Review, the authors discuss the current landscape of methodologies for using highly dimensional data resources and high-throughput biological measurements for combinatorial drug discovery.
